I don't see the "Best Dog" among the impressive assembly. No dog was cooler than Huckleberry Hound, the ever calm and always optimistic Hanna-Barbera classic!
As a "Son of the South" my own self, I always felt a kinship with Huckleberry, the coolest customer in the H-B gang.
Off-hand, I'd say Bandit, Lockjaw, DW's K-9, but above all them, I'd have to say mine...
Name's Daisy, an 8yr old german shorthair, always goes running with me, and YES, gives me those big sad eyes when she sees me put my uniform on because she know I'm leaving for a few days. Just the biggest baby in the world, but she loves a warm lap when I'm in the big mancave watchin' a movie drinking a microbrew..
Who wants to ever go to a theater when you have that setup at home......?
Oh, LOVE Brian from Family Guy. When I started watching years ago. I couldn't quite figure out whether or explain his ability to converse with humans, but he's like the most normal one of the bunch..
Also really enjoy Wallace's dog Grommit (especially when he's got his nose deep in books like 'Electronics for Dogs' or something (great deadpan)) and the pooch from 'How the Grinch Stole Christmas'.
I couldn't pick just one but Underdog & Snoopy are my favorite cartoon canines. I have fond memories of watching those old Underdog cartoons and with Snoopy, Charles Schultz added some great absurdist fantasy elements to the Peanuts gang. Even in Snoopy's fantasies, where he's the daring hero, he still couldn't take down that rascally Red Baron; the bad guy's always gotta be out there to keep the fantasy going.
Snoopy--art collector, with his Van Goghs in his doghouse; pilot on daring misions; and joyous dancer! Snoopy was also the first character I learned how to draw from memory, when I was sick at home from school. I had a Snoopy pillow, trashcan, and poster.
Scooby Doo...how about Hong Kong Phooey, number one superguy?...and Goofy was good in the cartoon about baseball.
I guess since Dino doesn't really count (grumble, grumble), I'd say Mr. Peabody - how can you not love a dog that has a time machine and his own pet human?
Along david_b's line, my true personal favorite is of course my own dog, Zita, a 6 yr-old mutt (there's some German Shepherd and Pit Bull mixed in there), who's temperament is a mix between Scooby Doo and Garm (the talking dog from Tolkien's "Farmer Giles of Ham"): she'd sell her soul for a snack, she's full of bluster at anything that runs away from her, but is actually rather frightened of 1. strange people, 2. dogs bigger than herself, 3. any animal smaller than herself, dog, cat or otherwise, that isn't impressed or intimidated by her initial salvo of barks and growls. In which case she turns around and tries to jump in my (or my wife's) arms, like Scooby.
But seriously, Brian Griffin is cool but he sometimes seems more like a person in a dog suit than an ACTUAL dog, you know? So I'd probably go with Snoopy.

Ah, the tech support behind all Wallace's inventions like the rocketship to the moon to find cheese, and still enjoys 'walkies' with his owner.
